I think it is general practice these days, for those using sequential media, to transfer in blastocyst media rather than in cleavage. I am sure the company providing your media will have guidelines as to how they think their media will best perform.

PESA/TESA sperm preparation is mostly depends upon the number of sperm obtained, mostly isolation of motile epididymal spermatozoa relatively straightforward,if large number obtained then mini density gradient is an effective method, if its low number simple wash will work. if its biopsy sample (percutaneous needle biopsy)then the testicular spermatozoa isolated by mechanical or enzymatic methods In enzymatic method collagenase enzyme is used to separate the seminiferous tubules, In mechanical method macerate the testicular tissue in culture medium by B D syringe

Re: Disinfection
The most easiest approach to disinfect the ambient of your lab. Is 75% homemade alcohol that is used at 15 minutes, let elements evaporate completely, before any pocedure performed.

Flushing Medium is a widely used solution and i am too using it regularly since last 8 years and i have no complaints with it. I have only one query, what is the utility of adding phenol red in a solution(HEPES based) with constant pH? Would you please answer?
