Re: Fertilization
Timing for fertilization check
Dear friends. I am very interested in this topic. I have done some research about this and here are some of my conclusions. Please comment freely.

1. Very rare 2 PN zygotes at 18 hours postinsemination can turn to 3PN or 1PN at 25 hours postinsemination (less than 2-3%)

2. Some 3PN zygotes can cleave directly to 3 cells and some can develop to nice embryos, BUT, do they do these at normal times or they can be selected out??

3. I am not aware of many published studies checking at 25 hours what happened to 3PN zygotes seen at 18 hours but in one I did found 70% of 3PN were still 3PN at 25 hours.

3. At what time do they cleave to 3 cells or 2 cells with an extrusion? I have found that the length of the cell cycle of 3PN zygotes is longer so many will be still be 3PN at the 25 hours check.

4. 2PN zygotes at the 25 hours check can form 4-cell embryos on day 2 so we can not distinguish them from the early cleavage ones, so this viality predictor is not useful if the embryos are cultured in groups.

5. At the 25 hours check if the zygotes is still at 2PN 19% reach blastocyst stage and have a 10% IR. If they are at pronuclear breakdown 26% blast rate with 22% IR and if at 2 cells stage 39% reach blast stage with 32%.

6. Next year we are going to check only at 25 hours and group the zygotes in 4 drops. (a:2PN, b: pronuclear breakdown c: 2 cells. d: other) and will choose embryos for transfer from groups c and b before group a. All group a embryos not selected for transfer will be left in observation to day 5.
