| I am working in bovine OPU IVP. and I have some problems with fertilization when checking cleavage rate at D2 after IVF where no more than 40% is achieved. I had checked fertilization medium ph at begining and ending (7.6 and 7.5) of 18 hs of IVF (TALP Fertilization medium), semen has been already used before and works well, incubator is pretty new but I dont really know how acurrate is CO2 injection and concentration, bicarbonate concentration in media could be not enough to equilibrate? any su..

No Cleavage after Fertilisation.

| Hi, I had this problem recently a couple of times, fert check at 18 hrs post ICSI showed 14 ova had 2pn but at 48 hrs only 7 had reached 4 cell stage, no cleavage in the rest, no pn either. I cultured the embryos further to 72 hrs, then 2 more embryos cleaved to 2 cell stage, no development in the rest. Other patients embryos which were cultured during the same time and in similar conditions progressed very well at 48 hrs. Checked everything from temp to Carbon dioxide saturation everything foun..

Effect of Vibration and Sound on embryo quality

| There is a very loud, strong compressor next to the IVF lab I am building. the walls vibrate and a drum like sound is clearly present. Besides issues with performing ICSI, are there data on levels of sound and vibration that would be detrimental to embryo quality and pregnancy rates.

Re: Effect of Vibration and Sound on embryo quality
Sound and vibration energies on in vitro embryo development An interesting question - there was some work years back - circa 1970's or so - related to the aerospace industry, on vibration and sound effects on in utero embryo development in mice (an maybe chick embryos) and I believe that there were some negative effects, though I can't say if the effects were fear induced, stress induced, or actual embryonic changes. There have been a number of similar studies with regard to gravity.
To your question, I have one abstract from the 2005 ESHRE meeting:
Effect of acoustic vibrations on mouse embryo development. O. Lopez-Rodrigo et al 2005 P-464 pg i167.
The conclusion states "Mouse embryos exposed to acoustic vibrations of moderate intensity within the range of audible frequencies do not show morphologic alterations, but their developmental rate appears to be faster than controls."
They evaluated two frequencies, but I'm not sure what the perceived physical effects would be - are these frequencies - 255.9 and 3465.5 HZ capable of being felt, e.g. physical vibration of the embryos as well, or is this just energy?
Perhaps this group from Spain will contribute more on their work.

Re: Slow development
Gold rule for IVF There are many possible reasons for slow development of embryos.
The room temperature in your lab is 26° C; did you try to reduce it to 23-24°C?
In one case an embryologist coming from outside measured the CO2 concentration in the incubator with a Fyrite and found it quite different from the display. Once adjusted the CO2 in the incubator all wen well. There are many things you should check. Anyhow go to the basics. The problem is often in front of your eyes. Moreover the embryologist should also have some knowledge of the stimulation protocols the clinician is using in every case and possibly be present at the ultrasound examinations during ovarian stimulation.

Re: Off gassing culture dishes
Different culture systems have to be gassesed experimenatlly. For Vitrolife 4 hours is enough without oil overlay.
We usually gas culture dishes minumun with oil 8 hours.
