| I have a Planer cell freezer. What type of "goblet" do labs use to put their straws in when freezing. Also, how many straws do you put per "goblet" so you can seed quickly. Do you seed both the upper and lower columns of media--above and below the embryo column?

Tough cytoplasm

| Hi, While doing ICSI, every now and then, we come across eggs with soft membrane. The oolema ruptures quite prematurely. So if we dont aspirate much cytoplasm, they would survive. But sometimes you happen to see eggs which would look normal when you inject the pip. inside. but when you aspirate, the cytoplasm is very tough and it doesn't rupture at all. In such eggs even slightest of aspiration leads to degeneration of egg. They are obviously fragile. What could be the reason? How should one handle such eggs?

Media usage Hi Samson,
It is recomended not to use different medias for the same set of gametes /embryos, they may have a different salt base which can cause metabolic shock to the embryos/gametes.
your incubator might be needing calibration at the operating temperature which should be kept unchanged (if its a TC sensor based), also, the supply pressure of co2 should remain unaltered.

No Problem! In our center we are used to incubate Hepes media tightly capped in CO2 incubators. The colour of the phenol red indicator does not change and fertilzation and cleavage rates were comprable with the incubation in the hot block. Probably, this depends on the type of the tube used in the incubation.
