Re: How to handle eggs with tough oolema with ICSI
Tough or Weak oolema membrane
Are you sure its a tough oolema? If you are seeing no breakage of the membrane upon aspiration of ICSI pipette, it is possible the oolema is too weak and has no resistance, which could explain the immediate degeneration. Try this, when you are positioning the injection pipette, take care to notice as it passes through the ZP to see if you get indentation of the oolema at all. If there is considerable difficulty in getting the pipette tip through the oolema then you have a tough oolema but if you see no signs of the needle 'punching' through the oolema then you have a weak membrane with no resistance. For a oocyte with a weak oolema, in my experience, use extreme care. As for oocyte with tough oolema, being a little 'rough' can help penetrate the membrane. By rough i mean a little 'stab' at the membrane, to prevent the membrane from squashing and moving the cytoplasm around which basically is the opposing force you are using to penetrate the oolema. Hope that sheads some light or at least promotes some further thoughts.
