Re: Best Vitrification Solution
Regarding the question about cryopreservatives for vitrification, PrOH is typically very toxic at concentrations necessary to vitrify if used alone. DMSO also can be toxic at high concentrations, and toxicity levels are often cell-type dependent. There seems to be an inverse correlation between ability to vitrify and toxicity, with the best vitrifying compounds, like PrOH also generally having the highest toxicity. Typically, combinations of compounds, used at lower concentrations, are used. However, I have seen PrOH showing extreme toxicity above 2M with mouse oocytes, and saw an identical phenomenon with human oocytes, although I did not have the resources at the time to do a controlled study to unequivocally determine if the cause of rapid cell death was the PrOH alone. I would avoid using PrOH to vitrify oocytes. To obtain more background information on this topic, search PubMed for publications by Greg Fahy and colleagues. If you can\'t get any PDFs easily, write to me and I\'ll e-mail some to you. - smullen@21cm.com
