| We have a lady 37 years old with 2 not very good cells for ICSI. After 2.5 days one of the cell divided in 2.( with no PN in the morning) I am wondering what might be the reason for this late dividing? Thanks

Incubators part three - relative humidity.
You want maximum humidity if you are doing embryo culture, even if you have oil overlays for your dishes, tubes and so forth. If you are doing open culture, you really - stress really - need to maintain maximum humidity.
First, older incubators, and a number of the new ones, rely on CO2 sensors that require maximum humidity for accuracy.
Second, anything placed into a dry incubator will be subject to dehydration at 37C - for example, open cultures (no oil overlay) will dehydrate quickly, raising osmolality to extremes and killing cells quite readily. And with oil overlay, I still worry about humidity - I\'ve seen the character of the oil change under dry, heated conditions, and that may translate to problems in the microdrops.
As for measuring humidity, why spend money on a tool to do this - 98%, 99%, etc.? The easy answer is to maintain at least one large pan of water in the incubator at all times, and if you are in a very dry environment, a second pan does not hurt.
I don\'t trust the manufacturer\'s recommendations when it comes to water pans - I have two newer incubators where the single pan provided might hold about 1 liter - so I purchased large rectangular glass pyrex baking pans that hold about 4 times the water, and use one of these in addition to the little pan supplied with the incubator.
Change the water every week to two weeks, but keep the pan or pans full. If your incubator has a door heating unit, you may not see condensation on the glass, but often you will. Condensation is a good sign that you are maximizing humidity.

I would also suggest that you take a very close look at the endotoxin load across your system, by retesting of products, media, and in particular, re-evaluation of the protein used in the culture, no matter what the certificates of analysis state.

Firstly, We used this test last year. my master thesis is about sperm DNA fragmentation. but I used this test diffirent research.We used to stain STAT III stain which contain Wrigt\'s Giemsa stain solution. It\'s work.detected by bright microscopy for counting halos.and was detected by according to literature.
Grade 1 : big halo (no fragmentation)
Grade 2 : medium halo (no fragmentation)
Grade 3 : small halo (fragmentation this step is complicated that is needed by experience)
Grade 4 : no halo (fragmentation)
Grade 5 : no halo and no stain or other (fragmentation)

Re: Gas Concentrations in Trigas Incubators used in IVF
As regards the triple gas mixtures we are totally at the mercy of suppliers. It on their assurance that we have to depend on. In a certain brand of incubators which strictly demand the triple gas mixtures, it would be like gambling with our IVF cycles. Though at times luck favours the brave but in two very prestigious labs in India, the results came crashing down thanks to the triple gas mixtures. Instead of us jumping into the mol bio and physico chemical jargon, we should nip the problem in the bud, else our 5 or 6% co2 are ideal.
