| What is the minimum volume of media for embryo transfer, below which the pregnancy rate drops? When somebody says "load embryos in as little media as possible", is there any lower limit?

Re: ph checking
I would like to know how does humidity influence the pH of the culture media used. We are using galaxy incubators for our embryo cultures and often i find the humidity fluctuating between 90-97 %? If the humidity keeps on varying in this range every now and then, how can you adjust the CO2 % to attain the best pH for embryo culture?
Shane
Re: ph checking
The humidity that you mentioned is enough. If you use culture under oil, humidity has secondary effects.
About PH, follow Thomas B. Pool\'s sugestion.Use a colibrated PH meter and adjust ph around 7.3, independent of C02 concentration on display/Fyrite or media provider.
I hope I helped you

Re: Re: ph checking
Thanks a lot. I think it will work. Which is the pH meter that you are using? Does it have atemperature probe attached to it? The article by DR. Thomas was excellent.
Thanks in advance.

Re: Loading embryo transfer catheter
I believe about 20 microliter medium plus air bubble (10-15 microliter medium)is used by most embryologists. How to load embryos with 0.4 microliter medium?
Weihua Wang
Re: Loading embryo transfer catheter
Hello. I read in one of the articles in Human Reproduction that the best pregnancy rates can be achieved when embryos + medium transferred into the womb is 30 - 40 microlitres. Correct me if I\'m wrong.
MayQ
Re: Loading embryo transfer catheter
0.4 microlitres is sufficient as we transfer into the uterus there is no need for external resource.

Re: Re: Total cleavage failure following conventional IVF
Fertilization checked conventionaly, 16-18hrs post-insemination for 2PN. 26hrs no cleavage noted when being checked for early cleaving embryos. Same status at 48hrs.
most appeared arrested at 2pn,some shrunk.
